If we have MBAM 2.5 installed on Windows 2012 server OS, can we in place upgrade WIndows OS to 2019?

Ok, sounds good. Yes, there's always that risk with an in-place upgrade. Better option may be to stand up the new one, patch fully, migrate the role / configuration and move on.

If you meant 2012 R2 then yes it is supported.

but a clean install, patch fully, migrate roles over is always recommended.
