You can set the 'base' features for Defender for Endpoint in:
https://security.microsoft.com > settings > endpoints > General:Advanced Features.
If you want more granularity you can use Intune - this includes folder exclusions.

When we onboarding my windows 2012 R2 to Security Microsoft Portal by installing windows defender for endpoint, this mean the server 2012 only report threat activity to the portal or this windows defender for endpoint can protect the server from virus? I can't see the defender for endpoint have GUI, so how i can configure the protection, whitelisting some folder etc.?
