I just discovered that all you need to do is use Expandable String Values (REG_EXPAND_SZ) instead of regular string values for the "NewSignature" and "ReplySignature" keys. This worked for me.

in our company we force to use specific signature generate by a script. To force it we add 2 regkey in HKEY_CURRENT_USER\SOFTWARE\Microsoft\Office\16.0\Common\MailSettings with 2 news value NewSignature and ReplySignature but it's not working anymoren, the signature seems selected in the signature menu (it's greyed out with signature name but it's not loaded when you click on "New message" as before in the old version (2019 and old version of 365). I think the issue coming after last update of office.
if i remove key from registry, it's working.

I understand that the signature can work normally when you remove the registry key, what are your concerns?

Do you mean that you want to make the specific signature as default signature when you send/reply a email?
If so, as i know, we can do that directly in Outlook desktop client, just refer to the below image. It's suggested that you could try it.
