Hi,
The OSDisk variable is used to tell your "Apply Operating System Image" task sequence step which drive to install the OS onto. The variable is only use in TS.
Upon the complete initiation of the full OS, it will reassign the drive letter.

i know the diskpart command and I know how I can use it. It was not my question how I can use diskpart.
My question was about TS and how I can use the letter drive and I have asked about two letter drive D: and E:
Considering your specific needs, I recommend you do not assign the drive letter to the drive E: (disk0-partiton5) at first.
Once the full OS is fully initiated, the second hard drive(disk1) should automatically get and hold the drive letter D:
Subsequently, you can utilize a script/TS to assign the drive letter E: to disk0-partiton5.
