I have checked C:\Windows\System32 for group policy folder to modify the gpo settings but did not find any folder related to Group Policy.
Please help me to solve this issue.

1,Based on my understanding , when you try to edit the GPO, the error happened, right?
2,All the gpos have the same issue or just one of them? Did all the DCS have the issue or just one of them?

Yes while editing the gpo I have faced the issue.

All the gpo have the same issue. Only one DCS have this issue.

I was trying to edit the gpo using Administrator.

Dcdiag /v >c:\dcdiag1.log
Repadmin /showrepl >C:\repl.txt

I would recommend you check if the problematic DC's sysvol can replicate from other DCs.
And if the clients have the access to the the problematic DC's sysvol by \dcname\sysvol
If the if the problematic DC's sysvol do have a problem, i would recommend you do a non-authoritative synchronization for the DC.

Before changes, backup the sysvol folder for all the DCs.

Sorry for the delay. I have performed non-authoritative synchronization for the DC but still I am facing the issue.
as per the above article, I have disabled SMB1 and SMB2 protocols in the server earlier. Later I have started facing the issue continuously on all the DC's.
I have modified the setting now to SMB1 as false and SMB2 as true. Now the issue seen for GPUPDATE is resolved.
I am not sure how it resolved but can you please explain how it is related to SMB protocols.
