Hello,
We use Office 365. When start to editing an excel spreadsheet "ctrl + ;" working as expected but after a while nothing happens when click "ctrl + ;" . All other shortcuts working as expected. We have to restart excel.

Did you use third-part keyboard in Windows?
I suggest you try to reset keyboard settings to default in Windows 10:

We changed the keyboard and reset keyboard settings but neither worked.

If this problem not related to keyboard, try to repair Office application and check the result:

Unfortunately we have already try that with no success....
