My desktop / HyperV host has 4 x 4K monitors. I want the guest to use 2 x 4K (2 full screen) + 1 x 1080P monitor (part of the third monitor). How can I accomplish this? Thank you

From your description, we find the issue is related to Hyper-v, I would remove the tag of "msc-virtual-machine-manager-hyper-v", which represents deploying Hyper-v in SCVMM. Thanks!

Does not explain though to use 2 full screen and 1 partial screen (1080p out of 4K) out of 4 x 4K monitors.
How would the RDP file be modified for that?

Where is the file to customize for that?
There is an rdp file in xml format, not sure how to modify it or if it supports all that rdp would support

I believe that the remote console only support 1920 x 1080 resolution.
Have you tried connecting with RDP instead. Maybe that will give you more options.
