If you mean an in-place upgrade, then no, that isn't supported.
Have you seen this thread about upgrading from BizTalk 2010 to 2016?
As long as you have a MSDN licence, you can use the BizTalk Developer edition, so no extra licence needed.
Try opening your BizTalk solutions in Visual Studio 2019 with BizTalk Visual Studio extension installed etc. and see if you can upgrade in one step.
