Can I "Upgrade" to Windows Pro from the MS Store from a vendor installed Winodws Enterprise?

Can I "upgrade" Windows ENT to Windows Pro by installing a download direct from the Microsoft Store, and purchasing the license direct from Microsoft? I currently have Pro (from the MS Store) installed but it will not activate with the Vendor ENT Levano license. I am left with a partly functional system. (Please see the comment below for background - Levano was no help)
