I no longer want to associate an old shortcut key combination (as in desktop 'Shortcut') but rather reassign the key combination to an entirely new shortcut. How do I do this ?

To reassign a keyboard shortcut from one desktop "Shortcut" to another, you must first clear it from the original file before Windows will let the new one use it. Windows typically won't allow two different shortcuts to share the same custom key combination.
Step 1: Remove the Old Shortcut Key
Step 2: Assign the Key to the New Shortcut
