SMTP Receive Error 421 Every 30 Seconds

Our SMTP receive log shows an error every thirty seconds. SMTP traffic appears to be flowing fine from on-premise to O365/Exchange Online, but some users periodically report the following error:

the server response was: 4.7.0 Temporary server error. Please try again later. PRX5.

I have searched the web and found that this type of error could be caused by a misconfiguration where SMTP emails are not received or sent at all. However, SMTP traffic is flowing as expected. Could there be a secondary listener that needs configuring? Any suggestions for resolving this issue would be appreciated.

    Thanks for posting your question in the Microsoft Q&A forum.

    To fix this issue, you can be done in 2 steps.

    Step 1: Add a custom receive connector to Office 365 IP for Port 25.

    Step 2: Bind the SSL Certificate to three receive connectors: default, default frontend, above-mentioned custom receive connector.

    2. We believe it's causing a periodic failure. When it does so, the sending application will not retry.

    We determined that if you disable the default Frontend receive connector for security reasons, you need to create a new receive connector for the server to use.

    1. Create a new receive connector
      1. Name: <Server name> - Loopback
      2. Type: Frontend Transport
      3. Authentication: Transport Layer Security (TLS)
      4. Permission Groups: Exchange servers
      5. Remote network settings: 127.0.0.1 ; <Server IP Address>
      6. Network adapter bindings: IPv4 25
