Hi there
I have a web application that is using "Microsoft Search Server 2010 Express" for searching contents of *.doc and *.pdf documents.
We are planning to upgrade Windows Server from WS 2008 to WS 2016.
And from the OS requirement spec for MS Search Server 2010 , I can see it is supported until 2008.
And also it is not very clear if MS Search Server 2010 Express will be supported after 13/10/2020.

Search Server Express goes out of support as of October 13th, 2020. You should instead consider deploying SharePoint Server 2019 for your search needs or if you have a Microsoft 365 deployment, look into Microsoft Search + Power BI Data Management Gateway + File Share connector.

Yes, MSSE was based on components of SharePoint Server 2010 search. SharePoint Server 2019 has significantly improved search capabilities. It will fit the needs you outlined.

But MS Search server 2010 Express was a free one without any license cost to it without installing Sharepoint Server 2010.

No, you need to properly license your users and servers with 2019. If your users are using Enterprise features, they each require a Standard+Enterprise CAL. Each SharePoint Server needs to be licensed with a Standard or Enterprise license.
Microsoft does not have any current free solutions for search outside of Windows Server indexing, which isn't quite the same.
