Is it safe to defragment an in-use file with Contig?

Greetings,
I'm wondering if I should expect issues when running Sysinternals' Contig on an SQLite database that's concurrently being written to and read from. I think I may be running into an issue with an extremely fragmented SQLite file and I'm wondering if it's safe to defragment the file while it's in use.
The system is running Windows 7 with NTFS.
