We have Azure SQL (IaaS) and the backup option is grayed out because we are using Ultra disks. In that case how we can configure SQL backup?

You can back up your Ultra SSD-managed disks using the Aure Backup Windows Agent as explained here.
You can also use third-party backup tools like Veeam Agent.
