In Windows 7, the actual startup sound is built-in and can't be changed. However, as a workaround, you can uncheck Play Windows Startup sound on the Sounds tab, and change the sound for the Windows Logon event in the list.

You may try this:
a. Open Audio Devices and Sound Themes by clicking the Start button, clicking Control Panel, clicking Hardware and Sound, and then clicking Sound.
b. Click the Sounds tab, and then, in the Program list, select the event whose sound you want to change.
c. Under Sounds, click the sound that you want to associate with the event. If the sound you want to use isn't listed, click Browse to locate it.

Yet, another reason why Microsoft doesn't understand what people want: simple customization.
The startup sound changer mentioned above has been replaced by a program called Winaero Tweaker. It's a portable program (no installation process) that works well, and does more than just change sounds. I just downloaded and ran it. Link below:
After all the messing about, I found the best way is to write a short and simple bit of code to play the sound, in VB6 or whatever, and put it in the startup folder. Even simpler, put the wav file in the startup folder, but you will have to close down Windows Media Player afterwards! Better and SAFER than messing with boot up dll's .
