If you restarted your PC, without resetting the router or the network settings, does it work temporary?
Your problem description matches the Microsoft bug in a big way,, which a they will be fixing in early April.

I am running Windows 10 and I keep losing internet connection and when I run troubleshooter I get error message 'Windows cannot automatically detect proxy settings', I then open network & connections select proxy and see that 'automatically detect settings' is turned off, when I select turn on I am unable to save the setting and once I close the settings page and recheck the setting has reset to off?
I have tried many different options I have found on web to try and clear this like going to internet options selecting connections and LAN settings & find that I am still unable to save the option to 'Automatically detect proxy settings'?
I am using a direct Ethernet connection and have tried reseting my router and resetting my Ethernet adapter this usually resolves problem but only until the next time I log on to PC
Any help would be appreciated

Do you use a direct internet connection or a proxy?
There is a known issue now in regards of Internet connection when you use a vpn or proxy.
Microsoft is working on a solution which should comes via the normal windows update channel.

In suggest checking for updates regularly now now.
