The problem isn't corrupted system files - the same thing happened to me. The update is good. The problem is that it broke something that these programs used to be able to do. They basically sit minimized and listen for a Function key to be pressed. At which point they send WM_LBUTTONDOWN and WM_LBUTTONUP messages to the currently active window at the mouse's current coordinates. All of that broke. The autoclicker apps only work now if they are the active window.
Overnight windows update broke all autoclick programs
My autoclick program worked fine yesterday, last night windows did a auto update to the OS and now it no longer works.
Ive tried several programs, ive tried compatibility mode, ive tried to run as admin, ive tried both together.
But after careful observation of the problem ive noticed what the culprit likely is.
Auto clickers are required to work when they dont have focus because they click other windows resulting in the autoclicker losing focus. but with this latest update windows no longer allows programs to 'click' while it no longer has focus. ive tested this by tabbing over to the auto clicker and it clicks just once and then stops when it loses focus from clicking on a window.
This seems like a 'safety' feature that windows implemented but it also seems poorly thought out given that it just broke every autoclicker.
Is anyone else having this issue? is there any way to fix this or get around this issue?

And if that is the case what does their support say?

2022-06-04T16:49:33+00:00 I tried GSAutoClicker and was previously using FreeMouseAutoClicker. both have the exact same problem.
This isnt the programs that stopped working, but the windows update overnight that broke them. i dont know how many such programs can be trusted so dont want to keep downloading new ones.

2022-06-04T17:51:23+00:00 Hi, I'm Douglas. Welcome to the community. I'm happy to help you today
Try performing a system restore to an earlier date where everything still worked.
Press Windows key+R to open Run and
type: rstrui.exe Press Enter.
This will open System Restore Click the Next button.
Choose restore point to an earlier date where everything still works.
Click Next and then Finish,
after that disable Windows Update updates for 7 days
Hope this information helps you, please post the results again. We are available
Yours sincerely,
Douglas.
