I believe AVD is pretty locked down.
You may need to power up an Azure VM in order to add custom ports.

How do I enable ports on Azure Virtual Desktop VM ? Should I just enable from VM side or should I enable anywhere else like host pool?

You should check the NSG (Network Security Group) at the NIC or Subnet level and open/close the ports
