The company has a physical machine named winserver2012R2, which is equipped with the super v service. There are many virtual machines in the super v, and now I want to upgrade this server2012R2 to server2022. I would like to ask if it is possible to upgrade by directly running setup.exe in the iso image of server2022. This way, the virtual opportunities under the super v in server2012R2 will not be affected before the upgrade?

Generally speaking, skipping one version is supported but, in all cases, an in-place upgrade is very risky move. A better option may be to export the VMs to removable, clean install the host, patch fully, add the hypervisor, import VMs, and move on.
