Are you connecting to Exchange server via https://localhost/ecp and getting the 500 error?
If yes, please try using another browser to see if this issue persists.
If you get the exact same 500 error message, please try creating a new mailbox via Exchange Management Shell and login https://localhost/ecp with the credentials of this new mailbox to see if the issue persists.
How to: Use the Exchange Management Shell to create user mailboxes
If the new mailbox also gets this error, please follow this link to recreate the ECP virtual directory via Exchange Management Shell:
