You need more information to analyze your problem, try using failed request tracing to see details about 500 error, this will generate detail log file, which will help you to identify the problem.

In IIS log I can see following 500 21 13 2

In the IIS manager, set the error pages to display detailed errors.

You should then get a page that looks like this.

What details do you get?

If you want us to help you, you're going to need to share details about how you've configured IIS and what you are browsing.

Does the default page display or is it just the contents of the directory that you moved?

Does Powershell give you the same error, or does it return the correct 200 status code? Can it access the default page?

Your question asks about a "virtual dir". Please explain what exactly you did. Did you define this directory to IIS with the Add Virtual Directory dialog? Is the physical directory on a drive that is local to the server that hosts IIS or are you trying to point IIS to some network share?
