Can Azure Document Intelligence automatically populate fields from an Excel or Word document, instead of requiring manual entry?

Are you trying to upload labels on your own in the custom model without using DI Studio to create it?

If yes, using the Document Intelligence Studio is the recommended method for label creation due to its proven quality and reliability. The labeling tool generates JSON files, including "fields.json" and ".labels.json".

While it is possible to create these files manually, it is not recommended because there is a significantly higher risk of introducing errors and inconsistencies.

You can try Auto label feature if that helps your case.
