If you are using SCCM, you can start from here to create a capture media:

Then create a task sequence to capture an OS:

I am using Rufus and ADK, PE and Sysprep. I was not sure what process was the easiest for custom deployments?

So it's not a question related to MDT or SCCM, personally I would recommend you try the steps to create a custom ISO image for mass deployment with a USB:

Prepare your lab environment by downloading the necessary media and installing the Windows ADK with Deployment Tools, USMT, and Windows PE options.

Create a bootable WinPE USB drive using the Deployment and Imaging Tools Environment and the makewinpemedia command.

Boot the reference computer into WinPE from the USB drive.

Use Sysprep to generalize the image and prepare it for capture.

Capture the image using DISM and save it to a WIM file.

Deploy the image to other computers using the Apply wim option in WinPE.
