I Have two DAG members in Exchange 2016 with 3 DB, MB01, MB02, and MB03.
MBO3 Active Copy of First Server gives below error in ECP:
"The Microsoft Exchange Replication service encountered an error while inspecting the logs and database for MB03\EXC01 on startup. Error: File check failed: Logfile 'G:\MB03-Log\E0300000004.log' was not found."
Is it an urgent problem? What must I do to solve this?
Are you running any anti-virus software on those servers? If so, make sure you are excluding the correct things

Hi,
Check the storage location, is the log file missing? Did you manually delete it?
Run the following command to check the mailbox database copy status and post the result:
Get-MailboxDatabaseCopyStatus -Identity DB1 | Format-List
Can you find any related error in Event log?

The log file "G:\MB03-Log\E0300000004.log" is not there really, it is lost. All filenames in that directory like "E03000007**.log" .

I am using the same antivirus on both Exc servers. These log directories were not excluded, but one of the log files has been deleted or lost anyway.
If that file deleted by antivirus or manually how can I correct the situation?
First:
Follow the doc and set the exclusions now before you do anything else.

Second: After you set the correct exclusions, remove the database copy on that server and reseed from another good copy from another mailbox server:
Remove-MailboxDatabaseCopy -Identity MB03\EXC01
Reseed:
Add-MailboxDatabaseCopy -Identity MB03 -MailboxServer EXC01 -ActivationPreference 1

Is there a risk of doing this process on Exchange? I am not so Experienced so should I be irritated?
The reseeding is very safe. You have redundancy.
Its a bigger risk to have the anti-virus installed on the Exchange Server and not set the exclusions, so its important to fix this as soon as possible.
