Cannot Update Path Environment Variable

I discovered that when I remove, or add an entry to path, or PATH, I no longer can run any of my libraries. For example... py, python, env, etc.
They all give me an error : '___' is not recognized as an internal or external command, operable program or batch file.

I then have to revert any change(s) I made to path, in order to get rid of those errors.
What causes this problem? Why can't I add, remove, or change entries in path without issues?
