Is it possible for in-place database partitioning?

Dear Sirs

We are planning to partition our big database into multiple partitions for better manageability. But the facing problems is when the database partitioning starts. The system would have to be stopped for a few hours, even longer.

is it possible to conduct the database partitioning while the system can still be accessed?

or the database partitioning can start for a few hours, then it can be pending until the next suitable time slot ?
