Is the Dump Stack file needed for windows to operate properly? I am noticing on various backups my external drive is giving me notices that two files could not be backed up and the Dump Stack file is one of them.

This is a system file; it is used as log file during dump creation when BSOD occurs.
If this file will not be included to backup, nothing harmful will happen. File will be re-created if needed.
User folder should be backup usually. What tool do you use for a backup?
You may ask Seagate support for a help about User folder.

Upon further view I notice what appears to be my User folder not been backed up which also seems to be setting in the C: directory. Would this be an issue?

Are you talking about file with .MDMP extension? If yes, this file is not needed for Windows, this file is created by SQL server. See some details at https://social.technet.microsoft.com/wiki/conte...
