You should disable the anti-virus completely and test.
Note that file-level anti-virus needs to have exclusions set:

I have installed a second Exchange 2016 server in an existing environment to create a DAG between the two servers but after installing the second server I have noticed the delay of emails that are stuck in the queue from the second server but if we disconnect the newly installed server, the emails are being delivered smoothly. I am requesting your help to figure out why emails are being first routed to the newly installed server and why they are delaying to be delivered.
I am getting "The periodic heartbeat to the primary server (Server name) Failed" from Application event logs.
Emails can be delayed between 1 to 5 hours without being delivered. Your help is highly appreciated, thank you.

tells me the server is getting blocked, or the transport service is down or the server itself is down.Which server is getting that error?
HAve you verified the transport service is up and running?
What does this show on the new server?
Get-ServerComponentState -id <NewServer> Test-ServiceHealthAnything in the event logs on the new server?
Message routing through the new server is expected however:

2021-06-14T14:57:27.147+00:00 If you are seeing this error on the NEW server and its referring to the old server (Server name), then you have a network or firewall issue.
That error is generated if it cant talk to other server's transport service. So its either down or the networking or firewall is causing issues.
Is the Windows Firewall enabled on these servers?"The periodic heartbeat to the primary server (Server name) Failed"

Thanks for the update.
I suppose that you didn't add the new server to the source server of the send connector which is used to route messages to the internet.If the new server failed to connect to the old server, it may fail to deliver messages.
