Dear Experts,
My issue is really strange. the OWA 2016 is deployed at client end.
When new email arrives it shows in the inbox. After some time (almost 20 to 30 minutes) it mark it as read without even reading the email.
Could anyone please help me to resolve the issue.

Sure its not another client or mobile device marking those as read?

Do you mean that you are working with owa based on exchange 2016 server? If so, according to my tests, there may be no option that could mark messages as read after 20-30 minutes, so agree with Andy, it's recommnended to check if you have logged into other clients first.
Also, based on my research, we could log mailbox access by mailbox owners, delegates (including administrators with full access permissions to mailboxes), and administrators by using mailbox audit logging, please kindly check it and hope that would be helpful to you: Mailbox audit logging in Exchange Server
