You should be able to upgrade from Home to Pro and when you run the installer file in the Windows 10 Home, it will give you an option for upgrade.
But take a note, you couldn't downgrade from Pro to Home.

issue with MAK Windows 10 PRO
I get 10 MAK WIN 10 pro in VLSC and i have WIN 10 home machines and i want to upgrade them , but, i can't activate in windows configuration, only formating with usb windows iso, and activating.
Is there a way to activate the windows HOME to PRO, without formating the PC?

The question is really can you upgrade Home to a Pro version using a Volume License Product key.
Open an Elevated CMD prompt, run cscript SLMGR /DLV all >licenses.txt
Make this available to download.
This is a list of the licenses that are on the Home edition which tells us which products keys can be used which are dumped into the license.txt file.Home is not a Qualifying OS for Volume Licensing as per this Licensing Brief.
Certainly going from Home to Pro retail will work.
