Actually, we don't know.
OneDrive must be on a drive that is using the NTFS filesystem.
Microsoft did not explain why OneDrive no longer supports non-NTFS file systems.

I am very interested in the "Resilient File System" (ReFS) and, to my surprise, found that after I reconfigured the "D:" drive on one of my computers to use ReFS instead of NTFS, "Microsoft OneDrive" would not allow me to use the "D:" drive. I received the following error message, which I typed from the attached screenshot.

Note that clicking on the error message dialog box's "Set up OneDrive" button had no apparent effect.
I have the following comments about the error message dialog box's body's message.
The following revised message includes corrections for my preceding observations.

OneDrive must be on a drive that is using the NTFS. To have OneDrive use a different location, click "Set up OneDrive," and point OneDrive to a directory which uses the NTFS. To use D:\ with OneDrive, you need to format it with NTFS and then click "Try again" to configure your account.
END REVISED MESSAGE
If "Microsoft OneDrive" supports ReFS, then the following two questions should become irrelevant, because the error message dialog box should be obsolete and no longer exist.
