Hi,
In This PC, USB was showing with two partitions. Then I did as below.
After creating a single partition, Diskpart is not able to format USB.
Then I did format from This PC as below.
Format was successful. Before creating bootable, I checked USB drive as below.
Then I tried to create bootbale USB from Rufus as below
And facing troubles as below
And I see USB drive as two partitions in This PC.
Please help me to understand and fix the issue. Moreover, pls let us know, what can be cause of this issue.

A couple of comments.
To format in diskpart you have to select the partition to format, you just have the disk selected.
Question, why are you formatting the disk in FAT32 and then formatting in Rufus as NTFS?
Is explorer open to Volume E?
Is there a CMD prompt open to Volume E?
Are there any other disk utilities that have Volume E Loaded?
before using fomat fs=fat32 quick, I used fomat fs=ntfs and fomat fs=ntfs quick but there was same issue.

As rufus is a third-party software, I recommend you to use the official media creation tool to create a bootable US for Windows 10.

I downloaded ISO from https://www.microsoft.com/en-in/software-download/windows10.
Most of Techies recommend Rufus for creating bootable. I also created previously many times.
As you said, As rufus is a third-party software, I recommend you to use the official media creation tool to create a bootable US for Windows 10.
In case of media creation tool, I see two options; upgrade existing Windows and download ISO.
Please let us know, what else should I use for creating bootable?

After error message, again I did same using Diskpart. Now I see only one partition.
Pls let me know, what I need to do?
Moreover, how do I know, if any issue in USB flash drive?
