We're seeing the same issue (intermittent 'Document not saved' errors when saving Excel files to a network drive), though we're not using Defender ATP.

Our environment:

  • Trend Micro WFBSS, cloud-based
  • Windows 10 1703 with Office 365 ProPlus version 1708
Microsoft 365 and Office | Excel | For home | Windows

    Hi Jim,

    We upgraded to Windows 10 1709, which we initially thought had resolved the issue. As with others who have reported this problem, it occurs with PDF documents, as well, so that should rule out Office. We don't run ATP, but tried this registry fix, anyway:

    • •            Key: HKLM\SOFTWARE\Policies\Microsoft\Windows Advanced Threat Protection\
    • •            Value Name: GroupIds
    • •            Value Type: REG_SZ (string)
    • •            Value: exclude_network_share

    This worked for about a day and the problem returned again, though with slightly different symptoms. Documents save, but TMP files are left behind and the network drive is sent into offline mode temporarily. If the user leaves the file open after a save, the drive stays offline for a few minutes, within which time another user can open and edit the same document, resulting in conflicts. It's a pain. 

    Anyway, I thought I would give an update on where we ended up before moving over to your linked thread in the Windows forums.
